目的 探討銅綠假單胞菌(PA)的密度感應系統(tǒng)(Qs)在氣道黏液高分泌中的作用。方法 60只大鼠隨機分為3組(野生型菌株PAO1組、QS基因突變型菌株PAO1-JP2組和對照組),分別將包被不同亞型PA的硅膠管置人大鼠一側主支氣管,建立慢性肺部感染模型,對照組則用生理鹽水浸泡硅膠管。28 d后處死大鼠,取肺組織和支氣管肺泡灌洗液(BALF)采用AB.PAS染色、ELISA和RT-PCR等方法檢測氣道內黏蛋白MUC5AC蛋白及mRNA的表達水平,比較野生型菌株PAO1和Qs突變型菌株PAO1-JP2對大鼠氣道黏液分泌的影響。結果 PAO1組較PAO1-JP2組氣道黏膜上皮的杯狀細胞數目顯著增多(P lt;0.05),對照組的杯狀細胞數目極少。PAO1組MUC5ACmRNA表達量較PAO1-JP2組顯著增多(P lt;0.05),對照組MUC5AC mRNA僅有少量表達。PAO1組BALF中MUC5AC蛋白分泌量明顯高于PAO1-JP2組(P lt;0.05)。結論 PA的QS重要基因缺損導致氣道黏液分泌的能力顯著下降。
