目的探討以殼聚糖為載體、采用乳化交聯(lián)法制備的bFGF殼聚糖微球體外釋放特性,為下一步實(shí)驗(yàn)奠定基礎(chǔ)。 方法應(yīng)用0.6%三聚磷酸鈉溶液作為交聯(lián)劑,1.5%殼聚糖溶液作為載體,采用乳化交聯(lián)法制備bFGF殼聚糖微球。激光粒度及Zeta電位分析儀檢測微球粒徑分布,掃描電鏡觀察形態(tài);ELISA法計(jì)算bFGF殼聚糖微球載藥量、包封率及體外釋藥規(guī)律。 結(jié)果bFGF殼聚糖微球粒徑為20.312~24.152 μm;掃描電鏡觀察顯示微球表面光滑圓整,無明顯孔隙,分布均勻,分散性好。載藥量和包封率分別為(7.57 ± 0.34)mg/g及95.14% ± 1.58%。bFGF殼聚糖微球可持續(xù)體外釋放bFGF 24 d;bFGF濃度隨時(shí)間延長逐漸升高,第24天達(dá)(820.45 ± 21.34)ng/mL;微球體外釋藥具有突釋效應(yīng),突釋率為18.08%,24 d累計(jì)釋放率為82.05%。 結(jié)論乳化交聯(lián)法制備bFGF殼聚糖微球操作簡便,微球表面光滑、分布均勻,分散性好,載藥量和包封率均較高,體外釋藥較穩(wěn)定且釋放率較高,是一種較理想的制備bFGF殼聚糖微球的方 法。
