目的探討As2O3對(duì)人乳腺浸潤性導(dǎo)管癌裸鼠移植瘤組織中NFκB p65、survivin及caspase3表達(dá)的影響。
方法復(fù)制人乳腺浸潤性導(dǎo)管癌裸鼠移植瘤模型,共22只,隨機(jī)分為3組: 對(duì)照組、順鉑組及不同濃度(1.5、3.0 mg/kg)的As2O3組(簡稱As2O3組)。用原位雜交法檢測survivin mRNA的表達(dá); 用免疫組化法檢測腫瘤組織NFκB p65、survivin及caspase3蛋白的表達(dá)。
結(jié)果人乳腺浸潤性導(dǎo)管癌裸鼠移植瘤中NFκB p65蛋白及survivin mRNA和蛋白表達(dá)的陽性細(xì)胞密度As2O3組明顯低于順鉑組和對(duì)照組(P<0.01),順鉑組也低于對(duì)照組(P<0.01); 而caspase3蛋白表達(dá)結(jié)果則相反(P<0.01)。NFκB p65蛋白、survivin蛋白及survivin mRNA陽性表達(dá)間相互呈正相關(guān),而三者的表達(dá)均與caspase3蛋白表達(dá)呈負(fù)相關(guān)。
結(jié)論As2O3可能通過抑制NFκB p65的活性抑制survivin,從而激活caspase3,誘導(dǎo)人乳腺浸潤性導(dǎo)管癌細(xì)胞的凋亡,且呈劑量依賴性。
